Insulin is key mediator in diabetes related hyperglycemia and hyperlipidemia. In the present study, we have investigated insulin related beneficial effects of swertiamarin using primary cultures of islets of langerhans and streptozotocin (STZ) diabetic rats. In STZ-diabetic rats, swertiamarin produced an increase in AUCinsulin which was not statistically significant. Treatment of beta islets of pancreas isolated from rats with STZ produced decrease in insulin level which was found to be increased by swertiamarin. Histology of pancreas from type 1 diabetic rats showed marked amelioration of damage as compared to diabetic control. littorale was collected during August – September at the end of flowering season. The plant was identified and authenticated by the taxonomist of Pharmacognosy and phytochemistry Department, B. V. Patel Pharmaceutical Education and Research Development Center, Thaltej, Ahmedabad, India and a voucher specimen (No. 2624) was deposited. The plant material was cleaned and dried in shade and stored at 25 oC. Preparation of aqueous extract of E. littorale One kg of shade-dried herb contaning all vegitative and reproductive parts of E. littorale was powderd in an electric grinder and boiled with 4 L water for 8 h and filtered. The filtrate was then concentrated to dryness under reduced pressure (yield 85.23 g). Extraction and Isolation of Swertiamarin Swertiamarin was isolated as described previously. Characterization of isolated compound was done by IR, NMR, mass spectra compared with reported data and Co-TLC of isolated and standard sample of swertiamarin [15]. Standard swertiamarin gifted by Prof. Fumikho Yoshizaki laboratory, Sendai, Japan. Chemicals STZ was purchased from Sigma (St. Louis, MO, USA).
