6.3.1. Preliminary batches for development of GRMDDS for baclofen Due to bioadhesive property of HEC, mucoadhesive tablets are expected to remain in the upper GIT and release its drug content for a long period of time, thus providing sustained therapeutic effect. Xanthan gum has been a dominant hydrophilic vehicle used in controlled release dosage forms because of its non-toxic nature, ease of compression, and capacity to accommodate high levels of drug loading. One of its most important characteristics is high swellability which exerts a significant influence on the release kinetics of an incorporated drug in it. Upon contact with water or biological fluid, the fluid diffuses into the device, resulting in polymer chain relaxation leading to volume expansion. The incorporated drug then diffuses out of the system. 6.3.2. Precompression parameters of all formulation batches Preliminary batches of BCLF mucoadhesive tablets were prepared by wet granulation method using mucoadhesive mucilage and rate controlling polymer xanthan gum, microcrystalline cellulose, dicalcium phosphate as filler, talc and magnesium stearate as glidant, lubricant and water as granulating fluid.…show more content…
