Mixed Unknown Bacteria Lab Report Biology 2460-002 Jamuna Kadariya November 24, 2014 Unknown number: thirty-four I. Abstract There are many types of bacteria and they produce different enzymes. However, some bacteria can produce same enzymes. In order to identify the particular enzymes produce by each bacteria specific test needs to be done. Among various types of enzyme producing bacteria, gram- negative and gram-positive bacteria are given priority in our experiment. There are four main types of Gram-negative bacteria and four Gram-positive bacteria including: Escherichia coli, Klebsiella pneumonia, Enterobacter aerogenes, Salmonella typhimurium. Bacillus Subtilis, Staphylococcus aureus, Staphylococcus saprophyticus and Streptococcus faecalis. The main purpose of this study is to identify the given mixed unknown bacteria by gram staining and a series of biochemical test. The spores test was negative because the dyed cells appeared pink with no green so it did not have an endospore to stain. The coagulase test appeared to be positive because when it places in a droplet of serum and watch for the clumping of cells, visible clumps appeared in 1 to 2 minutes. The MSA plate also showed positive results, in the presence of a mannitol fermenter, acid is produced which lowers pH enough to turn the indicator yellow. The SFA test results turned out to be negative meaning no color change in the purple media. The media could not utilize peptone and dextrose so enterococcus did not grow and media stayed negative. For the bile test, the iron salts and 6,7-dihydroxy-coumarin react and color change from brown to black, which means it is positive for the bile
