hdCK3mut is smaller than HSV-TK, which is beneficial in constructing therapeutic vectors that have size limitations. hdCK3mut is more sensitive than alternative PET reporters, allowing for increased signal at sites expressing hdCK3mut (26). The engineered substrate affinity of hdCK3mut is toward thymidine analogs (27, 28), while HSV-TK is engineered toward acycloguanosine substrates (29). In some chemotherapy and bone marrow transplant regimens, a continuous administration of acycloguanosine compounds is given as prophylactic antivirals, limiting the use of HSV-TK in these settings (18, 25). The use of hdCK3mut as a PET reporter would allow for the continuous treatment with such antivirals.
